Research on the Preparation of Bacterial Artificial Chromosome Library Genomic DNA

Abstract: This paper introduced a preparing method of genome DNA for a Huoyan goose and it laid a foundation for the constructing of bacterial artificial chromosome library .The genome DNA was extracted from a goose’s blood and digested by three restriction enzymes of Hind Ⅲ,EcoR Ⅰ and BamH Ⅰ. Meanwhile，different concentration gradients were set in order to get the optimal concentration of restriction enzyme.The results showed that Hind Ⅲ was the best enzyme and the optimal concentration of restriction enzyme was 40 U/μL. The genome DNA abstained by this method had a good quality and could be used to construct the bacterial artificial chromosome library.
