Isolation，Culture and Identification of Skeletal Muscle Satellite Cells in Chicken

Abstract: Chicken skeletal muscle satellite cells were obtained from the leg muscle tissue of chicken embryo by tissue separation. The cells were studied on passage，cryopreservation and resuscitation，and identified by specific gene expression assays, and a new method was established on isolation，passage cryopreservation and resuscitation of chicken skeletal muscle satellite cells. The results of specific gene expression showed that the cells could express specific marker genes Pax7，but not MyoD，Myogenin of the myogenesis genes of muscle satellite cell，which indicated that the satellite cells were of higher purity and didn’t differentiate into skeletal muscle cell.
